Uji Aktivitas Enzim Katalase (EC 1.11.1.6) Terhadap Variasi pH dan Suhu
Abstract
Catalase is one of the groups of enzymes as an enzyme that degrades hydrogen peroxide (H2O2), the overall reaction for catalase can be described as the degradation of two molecules of hydrogen peroxide into water and oxygen. The performance of the catalase enzyme can be affected by several factors, so a qualitative test of the activity of the catalase enzyme is carried out to determine the optimum performance of this enzyme. The method was carried out using samples of natural ingredients, namely chicken liver extract, chicken heart extract, papaya leaf extract and turmeric extract and with the addition of a 3% hydrogen peroxide (H2O2) solution. The test was carried out under neutral, acidic (plus HCl 1 M), alkaline (plus NaOH 1 M), low temperature conditions (immersed in ice water) and high temperature conditions (heated). The results were obtained by observing the number of bubbles produced as a form of enzyme activity, the catalase enzyme can work optimally at neutral and alkaline pH conditions while the temperature conditions so that the catalase enzyme can work optimally is at high temperatures. Both can be known from the number of bubbles produced when studied under each condition.
